Methods of assesment and treatment of coeliac disease

This diploma thesis deals with methods of determining and treatment of coeliac disease. In the theoretical part of the work sums up informations about coeliac disease - history, prevalence, etiopathogenesis, immunopathogenesis, possibilities of diagnosis, discusses about gluten-free diet, mentions legislation of the Czech Republic and the European Union and compares expensiveness of gluten-free diet and normal diet. The experimental part of this thesis compares PCR, ELISA and indirect immunofluorescence methods for assesment of determination of genetic predisposition and antibodies in coeliac disease (greatest emphasis was placed on the comparison of antibodies against gliadin and deamidated gliadin antibodies in IgA and IgG).
Diagnostika parazitárních onemocnění a úspěšnost vybraných terapeutických postupů u lichokopytníků a sudokopytníků

This dissertation dealt with the diagnosis and therapeutic procedures for the treatment of parasitic diseases in odd-toed and even-toed ungulates with selected anthelmintics. Between 2020 - 2023, 5.359 artiodactyls and 819 solipeds, namely cattle, sheep, goats, aurochs, mouflon, deer, bison, bison bonasus, horses, donkeys and ponies, were microscopically examined for the presence of nematodes, trematodes or cestodes in faecal samples in the Czech Republic. Molecularly, 76 cattle samples, 26 bison bonasus samples and 52 bison samples were examined for the presence of trematodes and C. daubneyi was detected. Parasitic infections were monitored in naturally infected individuals, and during the study no clinical signs suggestive of parasitemia were observed in any of the monitored individuals. The most parasitic infections were evaluated for H. contortus, O. ostertagi, M. benedeni, C. daubneyi and A. perfoliata. As part of the dissertation, a total of ten anthelmintics available in the Czech Republic were chosen, focusing on individual categories of parasitic infections in various animal species. Based on laboratory analyzes of LDA tests, the species of nematode was determined according to the developmental stage of the larvae, which helped to choose the appropriate anthelmintic during treatment. In conclusion, it can be stated that almost all anthelmintics have been shown to be effective in therapeutically treated farm animals.
Prevalence of kratom use among 8th and 9th-grade students in primary schools in the Nymburk district - a questionnaire survey

Background: Mitragyna Speciosa - a plant, more commonly known as kratom, that has a history and tradition in Southeast Asia for centuries. In recent decades, it has been gaining popularity worldwide. In addition to its benefits, which include energizing, pain and stress relief, and reducing opiate withdrawal symptoms, kratom use carries health risks along with the development of addiction. In the Czech Republic, kratom is not yet legislated, its use is not regulated and everyone, including children, has access to it. Aims: The aim of this study was to map the lifetime prevalence among 8th and 9th graders in the Nymburk district and patterns of use, their awareness in terms of effects, risks and availability, sources of information about kratom and attitudes towards it. Methods: The data were collected using a paper questionnaire in four primary schools in the Nymburk district. The schools were selected by random sampling. 287 pupils participated in the survey. Results: 17.8% of pupils had experience with kratom. Boys reported experience more often than girls. More than half of the respondents with experience have used kratom at most twice, got it for free from a friend and tried it out of curiosity. Prevalence of substance use among regular listeners of different music genres: a questionnaire survey

Background: Music is an essential part of human life. Substance use is often associated with music artists, but research has shown that the risk extends to their listeners. Studies show that the genres at risk include rock, pop, techno and rap. Listeners, especially during adolescence, may be influenced by this music and as a result be prone to various types of risky behaviour. Aim: The aim of the study was to determine the prevalence of substance use among listeners of different music genres, i.e. how substance use differs among listeners of different genres and to what extent they use substances. A further aim was to determine the CAGE screening questionnaire scores designed to assess the riskiness of drinking. Further, the research focused on the prevalence of lifetime dance drug use and subsequently in the nightclub setting. Methods: An online questionnaire inspired by the European Model Questionnaire was used for data collection, which contains a set of questions assessing the prevalence of substance use at lifetime, 12-month and 30-day intervals. Annotation: This thesis deals with dependency behaviour and internet addiction among youth between the age of 11 to 15. It is very comnon to use internet as source of information as much as source of fun among this popullation. But the modern problem with internet abuse is definitely recent uprise of numerous social networks. They became an essential part of common teenager's life where everyone can be anyone. The communication has no rules, it is easier and smiley faces are better to express emotions than do it in person. All these things are tempting for young people and make them to sign up to these networks and share thein private lives with almost the whole world. Not that having a profile like this is something wrong, but does it really have to be the main part how to communicate with friends and make new ones? Dealing with Gender-based violence at Universities : an example from the Faculty of Law of Charles University

This thesis deals with the issue of addressing gender-based violence in the academic environment at the institutional level as an emerging agenda of university administration. Gender-based violence takes various forms, ranging from normalized verbal expressions based on gender stereotypes (psychological violence), gender-based discrimination (economic violence), sexual harassment to sexual coercion or rape (sexualized violence). As such, gender-based violence constitutes an obstacle to the fulfilment of the functions of universities and has an increased negative impact on access to education for marginalised groups of students. Using a case study method, this paper examines what solutions to gender-based violence have been adopted at the Faculty of Law of Charles University as of June 2022 and how the institutional solutions are reflected by those involved in their development. The theoretical contribution of the analysis of the process of institutional norm creation through the perspective of socio-legal studies and feminist critique of law lies in the identification of several types of resistances to fulfilling the obligation to create a safe learning and working environment by regulating gender-based violence as a type of undesirable social behaviour through internal regulations. Prevalence of addictive substance use among persons diagnosed with ADHD

Background: Attention Deficit Hyperactivity Disorder (ADHD) is becoming an increasingly important research topic among adult ADHD clients. It is likely to be one of the risk factors for developing addiction. Most of the available research examines ADHD in individuals who are already being addressed as part of treatment in an addiction treatment service. Yet, these researches show that hyperactivity disorder does occur in persons with addiction issues. At the same time, hyperactive disorder is likely to be behind many of the complications in the progression of addiction interventions, increases the risk of early treatment drop-out, and places a greater burden on both clients and staff. Aim: The aim of this study was to determine the prevalence of substance use among adults diagnosed with ADHD, i.e. whether this specific group uses substances and to what extent. A complementary aim was to provide insight into what effects substance users reflect and what may be motivating their continued substance use. Bee trypanosomatids and their interactions with pollutants

Honeybees (Apis mellifera) are highly valued worldwide for their products, but also as pollinators of crops and wild plants. Due to the close proximity of bees in the beehive and their foraging habits, a large number of pathogens that weaken the bee colonies are spread both inside and between colonies. In recent decades, high annual losses of bees have been recorded, which still do not have a clear explanation. The sudden loss of bees (known as colony collapse disorder) is likely due to the interaction of multiple factors at once. A large part of the problem is attributed to viral diseases and some parasitic organisms; parasites of the Trypanosomatida group play an uncertain role. A member of this group, Crithidia bombi, is considered a problematic inhabitant of the digestive tract of bumblebees, which can cause serious complications. Close relatives, C. mellificae and Lotmaria passim, parasites of honeybees, are sometimes associated with their higher mortality. Pesticides, whose effects can be very toxic, have also come to the forefront in assessing bee losses, and most pesticides have therefore been banned in the EU.
